重慶研究院在污染物監(jiān)測、環(huán)境風(fēng)險(xiǎn)評(píng)估及污染物毒理機(jī)制上取得新進(jìn)展

近日,重慶研究院環(huán)境與健康研究中心團(tuán)隊(duì)在環(huán)境健康風(fēng)險(xiǎn)因子評(píng)估、環(huán)境污染物生物智能監(jiān)測、監(jiān)測POPs轉(zhuǎn)基因斑馬魚制作及污染物對(duì)斑馬魚的毒理機(jī)制分析方面取得最新進(jìn)展,相關(guān)研究成果分別發(fā)表在《Journal of Hazardous Materials》、《Environmental Pollution》、《Fish & Shellfish Immunology》、《Chemosphere》、《DNA repair》和《Scientific Reports》上。
隨著世界經(jīng)濟(jì)快速發(fā)展,空氣顆粒物 (PM)、納米材料、重金屬、有機(jī)污染物等大量排放進(jìn)環(huán)境中,嚴(yán)重威脅著生態(tài)安全和人類健康。環(huán)境與健康研究中心針對(duì)多環(huán)芳烴(PAHs)和二惡英 (TCDD) 暴露造成的健康問題,研制出具有生物監(jiān)測水體POPs的轉(zhuǎn)基因斑馬魚Tg (cyp1a: mCherry),其靈敏度可以達(dá)到0.1nM水平,為環(huán)境污染物POPs的智能檢測及預(yù)警奠定了堅(jiān)實(shí)基礎(chǔ) [1]。另外,中心團(tuán)隊(duì)利用Cas9基因編輯技術(shù)成功研制出mstna和mstnb基因敲除的斑馬魚模型,并首次探討了其在免疫調(diào)控中功能作用,為建立環(huán)境污染物的肌肉毒性模型奠定了重要基礎(chǔ) [2]。
最近,針對(duì)在發(fā)展中國家顆粒物排放造成的健康風(fēng)險(xiǎn)進(jìn)行評(píng)估,中心團(tuán)隊(duì)發(fā)現(xiàn)PM10和 PM2.5的污染誘發(fā)死亡率和呼吸道疾病,與常規(guī)生物質(zhì)燃料的依賴緊密有關(guān)[3];石墨烯和氧化石墨烯材料在醫(yī)學(xué)器材和日常生活用品中應(yīng)用廣泛,但是有關(guān)納米材料使用的安全問題存在潛在隱患。在納米材料的毒性評(píng)估方面,環(huán)境與健康研究中心針對(duì)氧化石墨烯(GO)在體內(nèi)和體外的毒性機(jī)理、DNA損傷與疾病的關(guān)系進(jìn)行深入研究 [4, 5]。研究發(fā)現(xiàn),GO暴露導(dǎo)致堿基切除修復(fù) (BER) 通路的 DNA 損傷反應(yīng),同時(shí)原子力顯微鏡 (AFM) 研究結(jié)果證明 HEK293T 細(xì)胞在暴露前的結(jié)構(gòu)物理變化[5]。此外,研究發(fā)現(xiàn),在革制品工業(yè)中帶來的重金屬污染導(dǎo)致工人的血液、尿液和毛發(fā)樣本的重金屬濃度顯著增高,且血液中超氧化物歧化酶 (SOD) 水平與 Cr、鎳含量呈顯著正相關(guān)[6, 7]。中心團(tuán)隊(duì)還深入研究鄰苯二甲酸酯 (DEHP)在干擾動(dòng)物雄性生殖內(nèi)分泌系統(tǒng)的類環(huán)境激素作用,發(fā)現(xiàn)長期暴露DEHP顯著抑制成年斑馬魚精子與卵母細(xì)胞的發(fā)生,誘發(fā)雄性精巢及其子代幼魚的DNA 甲基化水平改變[8]。這些相關(guān)研究對(duì)環(huán)境污染物毒理機(jī)制研究、環(huán)境健康的風(fēng)險(xiǎn)評(píng)估及污染物監(jiān)測技術(shù)的開發(fā)應(yīng)用奠定了基礎(chǔ),具有重要的理論和實(shí)用意義。
